Doo is well knit tale, the story begins with Vasanth (Sanjay) who works in the sub registrar office. He lives with his mom (urvashi) and hangs out with his friends (jeeva, jegan). His life takes a turn when he meets his school friend Swapna (nakshatra), a software professional.
In no time, they fall in love. However, as they get to know each other, things begin to change and they split.The story takes a turn with the arrival of Anu (sangeetha), a journalist and she takes a special position in Sanjay’s life. However, both Sanjay and Swapna are unable to come over each other’s absence.

Shiva and Gautam are buddies and HR professionals in the same firm. Gautam’s insatiable thirst for wooing women, leads him to abuse a bank marketing call that Shiva receives. Madhu, a miffed friend of the woman in the receiving end, starts to taunt Shiva by making prank calls which later blossoms into real romance just over the phone without having seen each other. When they feel the time is right to meet and communicate their mutual love, Madhu gets entangled in the bad books of a deadly organized crime syndicate. Her struggle to wriggle out of this with the help of Shiva and his friends is what forms the rest of the story.

The movie is set in Rajapalayam where Guru Moorthy (Sathyaraj), referred as ‘Kaalu’, is a bigwig. A moneylender by profession, he is a rich and influential man in the town.
Enters Eeeti (Sundar C),an individual with a never care, never mind attitude. He is always behind money and is ready to do anything for the currency. He meets Guru and gets close to him. They become buddies. Now comes Mahalakshmi (Saranya Ponvannan). Guru is against her and ensures that her peace of mind is shattered every time.

Thiruppaachi and Sengulam are neighboring villages with a history of enmity between them. Maayan (Manoj) and Machakanni (Riya Sen) meet on the day of her engagement and its love at first sight. Her marriage itself gets called off when the groom and his family insult Machakanni’s family and her romance with Maayan continues. But when her brother finds out about him, all hell breaks loose. The marriage is finalised but Maayan’s friends swear to unite the star-crossed lovers. There is also a subplot of Maayan’s aunt (Radhika) trying to get her daughter married off to Maayan.

Two music directors Bharathi (Vijayakumar) and Kannan (Nassar) used to be friends and composed music together. After Kannan has a fight with the director of their movie, Bharathi who feels embarrassed decided to split from Kannan and compose the music for the film by himself. He offers Kannan the chance to reconcile after this movie. Kannan rejects this and instead gives details about the fallout to the newspaper and Bharathi ends up losing the movie deal. A war begins and both music directors hate each other and can’t even stand to hear the other’s name. Bharathi is very successful while Kannan, who is basically unknown, drinks away his sorrows.

Achyut and Anand are brothers, Jyosthna is a tenant in their home. Both the brothers try impressing her.
