Frank Detomello is a slovenly zoo keeper, and simple father to the only young and bright girl named Shane, who catches a cold right after he swallowed a germ-ridden egg! The inside of his body is known as the “City of Frank”. An unlikely hero of a policeman white blood cell, by the name of Osmosis Jones, works as a member of Immunity cell. But with that germ-ridden egg comes a mortal danger: Osmosis discovers Frank has really contracted a villainous and black-hearted deadly virus known as Thrax who arrives and is plotting to ultimately overheat Frank’s body, killing him from the inside out! Thrax is motivated by trying to become the nastiest new virus, attempting to kill each new victim faster than the previous. His grandiose plan for Frank is death in 48 hours, breaking previous medical records! Meanwhile the pill that Frank took, Drix arrives in the body and covers Frank’s throat with ice to cover the irritation. Osmosis Jones becomes his newest partner.

Five assassins aboard a fast moving bullet train find out their missions have something in common.

The film starts in 1796, four years after the occupation of Malabar by the British East India Company and towards the end of the First Pazhassi Revolt (1793–1797). The revolt is led by Kerala Varma (Mammootty), the Raja of Pazassi Palace, of Kottayam house. The exploitation of the native Indian resources by the Company had culminated in popular revolts against its authority across the district. With the help of Kerala Varma Pazhassi Raja’s uncle Kurumbranadu ruler Veeravarma (Thilakan), who is jealous of Pazhassi Raja for his success and influence, and Raja’s old companion Pazhayamveedan Chandhu (Suman), the Company act against Pazhassi Raja. This forces Pazhassi Raja to escape to the forests of Wayanad. The tribal force led by Neeli captures Assistant Collector Thomas Hervey Baber (Harry Key) and his fiancée Dora (Linda Arsenio) in the jungle, in spite of his commander’s objection. The Raja treats Thomas Baber and Dora as his guest and releases them.

Desperate and terrified, the girls have only each other for support in this ordeal, till they reach their destination.

The film opens with the ageing gangster, Singaperumal, forcing himself on a young girl, Subbu. He is unable to perform, and vents his anger by slapping the helpless Subbu. Singaperumal is the grand don of crime in Madras. His lieutenant, Pasupathi, brings a proposal. A large stash of cocaine, worth about ₹ 2 crore, has entered the city. The guy bringing the stash wants to sell it for ₹ 50 lakh. Pasupathi sees the immediate easy profit, as well as the long term benefit (to control the cocaine market with such a large supply) and the recognition that they are able to pull off such a deal. The drawback is that the stash really belongs to their arch-rival Gajendran. Singaperumal knows that Gajendran is a vicious and unpredictable adversary, and the venture is risky and likely to become messy. He decides to pass. Pasupathi dourly suggests Singaperumal is getting old and rusty. He asks Singaperumal to loan him the ₹ 50 lakh so he can do the job himself. Pasupathi is prepared to face the risks, and in return for the loan, he offers Singaperumal a cut of the profits. Singaperumal agrees to the loan, but, instead asks Pasupathi to first get the stash and then think of the profit distribution. The money for the loan is brought to Singaperumal.
